Abstract

Arab chickens are a promising poultry breed due to their adaptability, disease resistance, and production potential. Enhancing reproductive efficiency through artificial insemination requires semen extenders capable of preserving spermatozoa viability during storage. This study evaluated the quality of Arab chicken spermatozoa diluted with different combinations of physiological saline and egg yolk extenders during short-term cold storage. Fresh semen was collected from three Arab chicken roosters aged 1–1.5 years and diluted at a ratio of 1:3 using five extender formulations: P0 (100% physiological saline, control), P1 (90% physiological saline + 10% egg yolk), P2 (85% physiological saline + 15% egg yolk), P3 (80% physiological saline + 20% egg yolk), and P4 (75% physiological saline + 25% egg yolk). Diluted semen was stored at 5 °C and evaluated at 0, 6, 12, 18, and 24 hours. Semen quality parameters included individual spermatozoa motility and plasma membrane integrity assessed using the hypo-osmotic swelling (HOS) test. The results showed a progressive decline in motility and plasma membrane integrity with increasing storage duration across all treatments. Statistical analysis indicated no significant effects of extender combinations on spermatozoa motility or plasma membrane integrity during storage, except at 0 hours for plasma membrane integrity. Although egg yolk supplementation up to 25% did not significantly enhance spermatozoa quality, the combination of 75% physiological saline and 25% egg yolk maintained spermatozoa quality comparable to other treatments during short-term cold storage. These findings indicate that physiological saline–egg yolk extenders are suitable for short-term storage of Arab chicken semen in artificial insemination programs.
