Abstract
Melitaea didyma is a butterfly that lives in grassland habitats that have sexual dimorphism. Female butterflies are bright orange with brown dotted on their wings both on the dorsal and ventral sides, while males are bright red with brown dotted. These butterflies are widely studied molecularly, because of their diversity in morphologic traits and are considered as species cryptic. This research is an exploratory study, which explores the genetic diversity of Melitaea didyma butterflies through analysis of the diversity of amino acid sequences of animal barcode regions from data available on Genbank. A total of 253 cytochrome amino acid sequences cytochrome C oxidase subunit 1 of the Melitaea butterfly were the alignment step by step according to their origin samples. The results of the alignment then re-aligned to compare and analyze the amino acid diversity with amino acid sequences from different origin samples. The different amino acids are analyzed their position and biochemical properties. The results showed that the amino acid sequence in the Melitaea didyma was less diverse. Of the 219 amino acids compared, only 3% (7 different amino acids). One amino acid is at the NH2 end, two in the internal loop, one in the external loop, and three in the transmembrane section. Amino acids in the internal and external loops are hydrophilic, while amino acids in the transmembrane are hydrophobic. Transmembrane amino acids are in the regions/zones M3, M4, and M5. Phylogenetic tree reconstructed based on 219 amino acid sequences of Melitaea didyma with the p-distance model, showed compatibility with the reconstructed phylogenetic tree using cytochrome C oxidase subunit I gene sequences; COI is an animal barcode sequence.
